SOP for receiving, storage and preparation of microbial culture media

1.0 Purpose:

To lay down the procedure for receiving, storage and preparation of microbial culture media.

2.0 Scope:

This procedure is applicable for receiving, storage and preparation of media in the microbiology laboratory.

Microbiologist :  To follow the entry and exit procedure in microbiology laboratory as per defined procedure.
Head-QC : To review and Implementation and ensure compliance of SOP.
Head-QA : To approve , implement and ensure compliance of SOP.

4.0 Procedure:

4.1 Receipt of media:

4.1.1 Microbial culture media must be procured from authorized agencies.

4.1.2 On receipt of media first check visually for following details i.e. cracked containers, frozen or melted media, unequal filling in container, change in expected color of media, these media must be discarded, documented & reported to vendor.

4.1.3 If media container pass in visual observation then record the following details i.e., enter the details such as name of medium, code number of the medium, date of receipt, quantity received, batch No./ lot No., expiry date, date of opening, container no. tested and date of release lot and give them individual bottle numbering.

4.1.4 A certificate of analysis should be received with consignment or available on the web site.

4.2 Storage of Media:

4.2.1 Store all the media in a cool and dark place at appropriate temperature. If any media requires specific temperature in the range (2 - 8°C), then it shall be stored in the refrigerator.

4.2.2 Label all the container with open date, expiry date, used up to date and container number.

4.2.3 Perform the growth promotion test, pH and pre-incubation check of the enriched media before using in microbiological analysis.

4.2.3 After passes of these test note down release date and now it can be use for routine use.

4.2.4 The date of release of other container of the same lot will be the date of the 1st container released.

4.3 Preparation of Media:

4.3.1 Ensure that the media stock is used on a first in first out (FIFO) basis or First Expiry First Out (FEFO) and that only one pack of medium is in use at any given time. On opening new pack enter the date of opening on pack and in record.

4.3.2 Weigh a required amount of dehydrated media and add it to the clean conical flask containing purified water. Gently mix it. In case of agar media slightly heat the media for dissolving of agar.

4.3.3 Check the pH of the media before sterilization and record it. Adjust the required pH using 0.1 M HCl or 0.1 M NaOH.

4.3.4 Distribute the media in the flasks or tubes as per the requirement. Cover the flasks and tubes with non adsorbent cotton plugs and wrap with butter paper or aluminum foil.

4.3.5 Autoclave the media at appropriate temperature, pressure and time as per validated autoclave cycle. (Generally at 121°C and 15 Psi pressure for 15 minutes.)

4.3.6 After completion of autoclave remove the media from autoclave and allow cooling up to 55°C. Pour it into petriplates (15-20 ml in each plate) or prepare slant (10 ml in each tube) under LAF.

4.3.7 Allow it to solidify. Check after pH and note down. pH after sterilization should be within the range for routine use of media.

4.3.8 Put all solid and liquid media for pre-incubation between 20-25°C. Use media as per validated use before time period.
